Description

A multi-step validation study was conducted to assess the use of digital PCR (dPCR) for detecting this mutation in biopsy tissues and urine samples. The dPCR assay proved specific, sensitive, and linear, with a prevalence of the V595E mutation in 22 urothelial carcinomas at 90.9%. When compared to histopathology, the diagnostic accuracy of using the BRAF mutation as a surrogate assay was 85.7%, with a positive predictive value of 92.3% and a negative predictive value of 80.0%. The findings from urine and biopsy tissue matched completely in all cases where both were analyzed. Combining the dPCR assay with urine sediment cytology significantly improved diagnostic accuracy in cases with clinical suspicion of malignancy. [Ref: Gentilini, F., Palgrave, C. PBMCs are used in antibody discovery due to their role in the immune response. Antibody-secreting cells (ASCs), a type of PBMC, are an excellent source of high-affinity antibodies with therapeutic potential. Millions of PBMCs can be isolated and screened to identify antigen-specific ASCs that secrete antibodies with desirable properties. One method involves microfluidic encapsulation of single ASCs into an antibody capture hydrogel, followed by antigen bait sorting using flow cytometry, enabling the rapid discovery of monoclonal antibodies. These antibodies can then be further characterized for their binding affinity and neutralizing capacity, making PBMCs a valuable resource for developing antibody-based drugs and studying immune responses.
