Description

This study aimed to establish two- and three-dimensional primary cell lines from oral canine melanomas using fine-needle aspiration, a minimally invasive sampling method. Samples were collected from six dogs, including four primary oral melanomas and five lymph node metastases. The cells were processed into single-cell suspensions and seeded in flasks or formed into organoids using Matrigel. Characterization was performed using flow cytometry with antibodies against Melan-A, PNL2, and Sox-10. This approach provides a non-invasive means to obtain cell samples, particularly beneficial for patients unsuitable for surgery, and enables the creation of in vitro models for comparative studies in mucosal melanoma oncology. Notably, this is the first study to establish neoplastic primary cell cultures from dogs using fine-needle aspiration. PBMCs are used in antibody discovery due to their role in the immune response. Antibody-secreting cells (ASCs), a type of PBMC, are an excellent source of high-affinity antibodies with therapeutic potential. Millions of PBMCs can be isolated and screened to identify antigen-specific ASCs that secrete antibodies with desirable properties.
